I have a 79 Trailfie 340 and it takes about ten to fifteen pulls to get it started after it sits for a few days.I have resealed the engine and pressure tested it and all was good,cleaned the carb three or four times and all should be good,replaced fuel lines and check valve in the tank and replaced the fuel pump with a known good one.If I squirt a little fuel down the plug holes then it starts and runs on the first or second pull every time.It only does this after a few days of no use,if I ride it every day then it will start from cold with the enrichment circut in about two to three pulls and once it is warmed up it will start in a half a pull.

Verify your choke adjustment first as they can be finicky. If it runs and idles fine it is probably not in the carb. It sounds to me like a soft fuel pump or a bad check valve in the tank. The prime is being lost while setting is my guess. I just resealed a 80 trailfire 340 and that starts in 1 or 2 pulls every time. hope you figure it out and i hope this helps.
